Research: Work in the Tibbetts laboratory is aimed at understanding key signal transduction pathways that control cellular responses to DNA damage induced by environmental insults such as UV light and gamma-radiation. The bulk of this work is directed toward the related DNA damage-response kinases ATM (ataxia-telangiectasia) and ATR (ATM-Rad3-related). ATM and ATR, and their orthologs in lower eukaryotes, function as apical DNA damage-signaling kinases that regulate diverse cellular processes including cell cycle checkpoint activation, DNA repair, gene transcription, and apoptosis. ATM and ATR have both been implicated as tumor suppressor genes that are mutated in human cancer. In addition, mutations in ATM are causally associated with ataxia-telangiectasia (A-T), a syndrome of cancer, neurodegeneration and premature aging that affects approximately 1 in 40,000 births. ATM is a well established tumor suppressor gene and its mutation has been strongly linked to sporadic and familial cases of breast cancer. ATM research is highly relevant for understanding the impact of environmental DNA-damaging insults on cellular physiology.
