Research as of Spring 2008
In the absence of Wnt, beta-catenin is recruited into a destruction complex leading to its ubiquitination and degradation by the proteosomes. The on-state of Wnt signaling results in post-translational stability by degradation of this complex. Accumulation of beta-catenin in the nucleus activates transcription of beta-catenin/TCF target genes. Our lab recently found a target gene for beta-catenin namely CRD-BP. This CRD-BP binds and stabilizes mRNA of several genes (including c-myc and b-TrCP). Studies have shown that a third of the melanomas have upregulated Wnt expression and NF-kB is activated in most epithelial tumors. My immediate aim is to assess the role of CRD-BP in melanoma development. To this end, I am planning to analyze the expression of CRD-BP in primary tumorigenic properties of melanoma cell lines.

My research also includes the study of a chemopreventive agent Lupeol and its effect on melanomas. We plan to expose various melanoma cell lines to this agent and study the expression of CRD-BP and various other proteins.
